چکیده

Protein A (SpA) is a component of the cell wall of Slop, lylococcus aureers. SpA is unique in that it combines with the Fc portion of immunolgobulins from many speciesi,2) and has been used in immunochemical studies3,`>. Another distinctive f ature of SpA is its potent mitogenic activity on lymphocytes from several species, including hurnans5,iO}. SpA also exerts a mitogenic activity on both T and B cells 6,8,iO}. Moreover, SpA is a striking contrast to other polyclonal B cell activators (PBAs) because it combines with the Fc portion of immunoglobulins. It is thus likely that SpA reacts with cell surface immunoglobulin receptors and in turn delivers

Construction of a human recombinant polyclonal Fab fragment antibody library using peripheral blood lymphocytes of snake bitten victims

Human snake bitten poisoning is a serious threat in many tropical and subtropical countries such as Iran. The best acceptable treatment of envenomated humans is antivenoms; however they have a series of economic and medical problems and need more improvements. In this study a combinatorial human immunoglobulin gene library against some of Iranian snakes venoms was constructed. Total RNA prepare...

Pathogenesis of B-cell superantigen-induced immune complex-mediated inflammation.

Staphylococcal protein A (SpA) is representative of a new class of antigens, the B-cell superantigens (SAgs). These antigens bind to the Fab regions of immunoglobulin molecules outside their complementarity-determining regions. SpA, the best-studied B-cell SAg, reacts with the Fabs of most VH3+ immunoglobulins, which are expressed on 30 to 60% of human peripheral B cells. Therefore, B-cell SAgs...

The Sbi Protein Contributes to Staphylococcus aureus Inflammatory Response during Systemic Infection

Staphylococcus aureus is an important human pathogen that causes infections that may present high morbidity and mortality. Among its many virulence factors protein A (SpA) and Staphylococcal binding immunoglobulin protein (Sbi) bind the Fc portion of IgG interfering with opsonophagocytosis. We have previously demonstrated that SpA interacts with the TNF-α receptor (TNFR) 1 through each of the f...

Inhibition of IL-13 by Antisense Oligonucleotide Changes Immunoglobulin Isotype Profile in Cultured B-Lymphocytes

The link between IL-13 and bronchial hyper-responsiveness has brought this cytokine as a potential therapeutic target for asthma and allergic diseases. At the present study, we address the role of B cell derived IL-13 in the IgE and other immunoglobulin development. Antisense oligo for human IL-13 m-RNA was used to study IgE down regulation. Human B-lymphocytes were purified by positive selecti...

Lactate dehydrogenase enhances immunoglobulin production by human hybridoma and human peripheral blood lymphocytes.

Lactate dehydrogenase (LDH) derived from rabbit muscle enhanced IgM production by human-human hybridoma HB4C5 cells 12.4-fold at 320 mug/ml under serum-free conditions. LDHs from pig muscle and pig heart also accelerated IgM production 8.4- and 6.4-fold, respectively. The immunoglobulin production stimulating activity of LDH was not accompanied by activation of cell proliferation. LDH from rabb...
